4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. Goat anti-rabbit IgG-biotin (HA1101) at different dilutions were used in 5% NFDM/TBST at 4℃ overnight. Streptavidin-HRP Secondary Antibody at 1/20,000 dilution was used for 1 hour at room temperature.
HA1101_2.jpg Fig2: HA1101 was tested by direct ELISA, where wells were coated with serially diluted rabbit/mouse/human IgG (1,000-31.25 ng/mL, 4℃, overnight), blocked with 1% BSA (37℃, 1 hour). HA1101 (1/10,000 dilution; 37℃, 45 minutes) was added and detected by Streptavidin-HRP (1/2,000 dilution; 37℃, 30 minutes). Signal was developed by TMB substrate. Data from duplicates; +/- SD.

Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 15 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Rabbit anti-SP1 antibody (ET1702-02) at 1/100 dilution in 1% BSA in PBST overnight at 4 ℃. Goat Anti-Rabbit IgG (biotin-conjugated, HA1101) was used at 1/200 dilution. Streptavidin-FITC was used as the secondary antibody at 1/1,000 dilution. Nuclear DNA was labelled in blue with DAPI.
