Background: Receptor activator of nuclear factor kappa-B ligand (RANKL), also known as tumor necrosis factor ligand superfamily member 11 (TNFSF11), TNF-related activation-induced cytokine (TRANCE), osteoprotegerin ligand (OPGL), and osteoclast differentiation factor (ODF), is a member of the TNF superfamily. RANKL also has a function in the immune system, where it is expressed by T helper cells and is thought to be involved in dendritic cell maturation. The murine TRANCE cDNA encodes a type II transmembrane protein of 316 amino acids with a predicted cytoplasmic domain of 48 amino acids and extracellular of 247 amino acids. Mouse and human TRANCE share 85% amino acid sequence identity. TRANCE is primarily expressed in T cells and T cell rich organs, such as thymus and lymph nodes.
