Western blot analysis of MTBR-TAU 243 lsoform 2N4R(236-250) on different lysates with Rabbit anti-MTBR-TAU 243 lsoform 2N4R(236-250) antibody (HA724309) at 1/5,000 dilution. Lane 1: Mouse brain tissue lysate Lane 2: Mouse spleen tissue lysate (negative) Lane 3: Mouse cerebellum tissue lysate Lane 4: Mouse testis tissue lysate (negative) Lane 5: Rat brain tissue lysate Lane 6: Rat spleen tissue lysate (negative) Lane 7: Rat cerebellum tissue lysate Lane 8: Rat testis tissue lysate (negative) Lane 9: Pig brain tissue lysate Lane 10: Pig spleen tissue lysate (negative) Lysates/proteins at 20 µg/Lane. Observed band size: 50-80 kDa Exposure time: 6 seconds; ECL: K1801; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (HA724309) at 1/5,000 dilution was used in primary antibody dilution (K1803) at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature. Negative control : Spleen, Testis (PMID: 24309898, PMID: 24386422) |

MTBR-TAU 243 lsoform 2N4R(236-250) was immunoprecipitated from 0.2 mg mouse brain tissue lysate with HA724309 at 2 µg/10 µl beads. Western blot was performed from the immunoprecipitate using HA724309 at 1/5,000 dilution. HRP Conjugated Anti-Rabbit IgG for IP Nano-secondary antibody at 1/5,000 dilution was used for 1 hour at room temperature. Lane 1: Mouse brain tissue lysate (input) Lane 2: HA724309 IP in mouse brain tissue lysate Lane 3: Rabbit IgG instead of HA724309 in mouse brain tissue lysate Blocking/Dilution buffer: primary antibody dilution (K1803) Exposure time: 7 seconds; ECL: K1801 |

Fig3:
Immunoprecipitation (IP) 50 ng of full-length TAU isoform 2N4R protein (1-441 aa) and 200 ng of truncated TAU isoform 2N4R protein (1-256 aa) were incubated with 0.2 mg of negative tissue lysate from mouse spleen, along with HA724309 at 2 µg/10 µl of beads. Western blot was performed from the immunoprecipitate using HA724309 at 1/5,000 dilution. Anti-Rabbit IgG for IP Nano-secondary antibody (NBI01H) at 1/5,000 dilution was used for 1 hour at room temperature. Lane 1: TAU isoform 2N4R (full-length protein, amino acids 1–441) and TAU isoform 2N4R (truncated protein, amino acids 1–256) were spiked into negative tissue lysate from mouse spleen (input) Lane 2: HA724309 IP in TAU isoform 2N4R (full-length protein, amino acids 1–441) and TAU isoform 2N4R (truncated protein, amino acids 1–256) were spiked into negative tissue lysate from mouse spleen Lane 3: Rabbit IgG instead of HA724309 in TAU isoform 2N4R (full-length protein, amino acids 1–441) and TAU isoform 2N4R (truncated protein, amino acids 1–256) were spiked into negative tissue lysate from mouse spleen Exposure time: 16 seconds Blocking: 5% NFDM/TBST, 1 hour at room temperature Primary dilution: HA724309, 1/5,000 in primary antibody dilution buffer (K1803), 2 hours at room temperature Observed band size: 50-80 kDa |

Fig4:
Flow cytometric analysis of EL4 (left, negative) and Neuro-2a (right, positive) cells labeling MTBR-TAU 243 lsoform 2N4R(236-250). Cells were fixed and permeabilized. Then stained with the primary antibody (HA724309, 1/1,000) (red) compared with Rabbit IgG Isotype Control (green). After incubation of the primary antibody at +4℃ for an hour, the cells were stained with a iFluor™ 488 conjugate-Goat anti-Rabbit IgG Secondary antibody (HA1121) at 1/1,000 dilution for 30 minutes at +4℃. Unlabelled sample was used as a control (cells without incubation with primary antibody; black). |
