Immunoprecipitation (IP)
MTBR-TAU 243 lsoform 2N4R (225-242) was immunoprecipitated in 0.2 mg mouse brain tissue lysate with HA724446 at 2 µg/10 µl beads. Western blot was performed from the immunoprecipitate using HA724446 at 1/10,000 dilution. Anti-Rabbit IgG for IP Nano-secondary antibody (NBI01H) at 1/5,000 dilution was used for 1 hour at room temperature. Immunoprecipitation (IP)
20 ng of full-length TAU isoform 2N4R protein (1-441 aa) and 200 ng of truncated TAU isoform 2N4R protein (1-256 aa) were incubated with 0.2 mg of negative tissue lysate from mouse spleen, along with HA724446 at 2 µg/10 µl of beads. Western blot was performed from the immunoprecipitate using HA724446 at 1/5,000 dilution. Anti-Rabbit IgG for IP Nano-secondary antibody (NBI01H) at 1/5,000 dilution was used for 1 hour at room temperature. Lane 1: TAU isoform 2N4R (full-length protein, amino acids 1–441) and TAU isoform 2N4R (truncated protein, amino acids 1–256) were spiked into negative tissue lysate from mouse spleen (input) Lane 2: HA724446 IP in TAU isoform 2N4R (full-length protein, amino acids 1–441) and TAU isoform 2N4R (truncated protein, amino acids 1–256) were spiked into negative tissue lysate from mouse spleen Lane 3: Rabbit IgG instead of HA724446 in TAU isoform 2N4R (full-length protein, amino acids 1–441) and TAU isoform 2N4R (truncated protein, amino acids 1–256) were spiked into negative tissue lysate from mouse spleen Exposure time: 6 seconds Blocking: 5% NFDM/TBST, 1 hour at room temperature Primary dilution: HA724446, 1/5,000 in primary antibody dilution buffer (K1803), 2 hours at room temperature Observed band size: 50-80 kDa |
